When Amazon updated its Dietary Supplement Verification Program (DSVP) requirements in 2024, the notices went out through Seller Central compliance alerts — not press releases. Brands that missed the update found out the harder way: listing holds, suspension notices, and urgent emails asking for documentation that their existing certificates of analysis couldn’t provide.

The core change wasn’t complicated, but it had serious downstream effects. Amazon now requires that COAs for dietary supplement products come from ISO/IEC 17025-accredited analytical testing laboratories. Not a third-party lab. Not a contract manufacturer’s in-house quality department. An independently accredited facility whose scope of accreditation covers the specific tests performed. For Midwest supplement brands — many of whom had been operating with overseas supplier COAs or generic testing documentation — this created an immediate gap between what they had and what the platform demanded.

Here’s what the 2026 requirements actually look like, and how to close that gap efficiently.

What Amazon’s Verification Program Requires in 2026

Amazon’s DSVP operates in tiers. What they ask for depends on your product’s category, its ingredient history, and whether your listing has previously triggered a compliance flag. At the baseline, every supplement seller should be prepared to produce a COA from an ISO/IEC 17025-accredited analytical testing laboratory covering four areas:

Identity verification. For botanical ingredients, this means a chromatographic or molecular method — HPTLC, DNA barcoding, or both. Organoleptic assessment (look, smell, taste) does not satisfy this requirement under Amazon’s program or under 21 CFR 111.75(a)(1)(i), which mandates identity testing on each incoming lot of dietary ingredient.

Potency and label claim accuracy. Quantitative assay results that demonstrate your declared amounts are within the acceptable tolerance window — typically ±20% for dietary supplement claims under DSHEA. This requires an HPLC or UV-Vis assay against a validated reference standard, not a calculation based on formulation records.

Contaminant screening. At minimum: a heavy metals panel covering lead, cadmium, inorganic arsenic, and mercury, tested against USP <232>/<233> permissible daily exposure (PDE) limits. For categories with documented contamination histories — herbal powders, traditional botanical extracts, mushroom products — Amazon may also request pesticide residue data.

Microbiology. USP <61> and <62> results showing total aerobic microbial count (TAMC), total yeast and mold count (TYMC), and absence of specified pathogens including Salmonella spp., E. coli, and Staphylococcus aureus. Standard acceptance criteria for oral dietary supplements require TAMC ≤ 10³ CFU/g and TYMC ≤ 10² CFU/g.

What Amazon does not accept: supplier-issued COAs from a manufacturer’s internal lab, COAs from non-accredited testing services, or documentation that references test methods without specifying the analytical technique. If your paperwork can’t be traced to an accredited facility with a verifiable scope of accreditation, expect a compliance hold.

The Four Tests That Drive Most Listing Holds

Looking at the categories of incoming samples from brands dealing with Amazon compliance issues, the same four failures come up repeatedly — and three of them aren’t what most brands expect.

Botanical identity (HPTLC + DNA barcoding). This is the most commonly failed, and the most frequently overlooked. Brands that purchase raw material from an overseas supplier and private-label under their own name tend to pass through the supplier’s COA without independent verification. HPTLC chromatographic fingerprinting remains the gold standard for botanical identity under USP guidelines — it produces a visual fingerprint of phytochemical composition that can be compared to reference standards. DNA barcoding adds a molecular layer, useful for detecting species substitution even in powdered or processed material where morphological cues are gone.

Heavy metals via ICP-MS. Inductively coupled plasma mass spectrometry is the preferred method for elemental impurity analysis because it detects trace metals at parts-per-billion sensitivity. The USP <232> oral PDE limits for the four most common problem elements are: lead at 5 μg/day, cadmium at 0.5 μg/day, inorganic arsenic at 15 μg/day, and mercury at 15 μg/day. Botanicals that are extracted, concentrated, or spray-dried can push above these thresholds even when the raw plant material is borderline. Rice protein, Ayurvedic blends, and chlorella-based products have historically presented the highest risk in this category.

Potency assay. A label declaring “600 mg ashwagandha root extract standardized to 5% withanolides” is a two-part claim: the extract mass and the withanolide concentration. Both need analytical confirmation. HPLC with UV detection against a validated withanolide reference standard is the standard method. If results show 4.1% withanolides instead of 5%, you have a label accuracy violation regardless of how precisely the formula was assembled. Amazon’s verification team checks label claims against assay results, and a ±20% miss triggers a hold.

Microbial enumeration (USP <61>/<62>). Amazon requests microbiology results more often for powdered herbals, probiotics, and blended products with multiple plant ingredients. Failing USP <62> — presence of a specified pathogen — is an automatic product hold. Failing TAMC or TYMC limits is more commonly a supplier quality issue that surfaces when raw material is poorly stored or cross-contaminated during processing. Either way, a 5-day incubation window means this test is often the rate-limiting step in your turnaround time.

Why Your Lab’s Location Affects Your Compliance Timeline

Here’s an operational reality that doesn’t come up enough in the testing discussion: every day your sample spends in transit is a day that testing hasn’t started. A Midwest brand in Chicago, Indianapolis, or Columbus shipping botanicals to a coastal lab adds two to four days of ground transit each way. That’s four to eight days of delay before any analytical work begins, and the same again before results arrive.

For brands managing an active Amazon compliance hold — which typically carries a 72-hour response window before escalation — that transit gap can be the difference between a resolved hold and a suspended listing.

How to Build an Amazon-Compliant Testing Dossier from Scratch

If you’re rebuilding after a compliance hold — or auditing proactively — here’s a five-step process that works.

Step 1: Audit your current COA inventory. List every active supplement SKU and its primary botanical ingredients. For each COA on file, identify the issuing lab and verify whether it carries ISO/IEC 17025 accreditation with a scope that covers the tests performed. Accreditation certificates are public record — they’re searchable through A2LA, Perry Johnson Laboratory Accreditation (PJLA), or the relevant national body.

Step 2: Risk-rank your product line. Not all products carry equal compliance risk. Prioritize: anything with botanical raw materials sourced from South Asia, Central Asia, or China; any high-volume SKU where a hold would have significant revenue impact; and any product that makes a specific quantified claim (e.g., “standardized to X% active compound”). Start testing there.

Step 3: Scope your testing package correctly. For each product, the minimum Amazon-defensible package typically includes: botanical identity (HPTLC ± DNA barcoding), heavy metals panel (USP <232>/<233>), microbiology (USP <61>/<62>), and a potency assay for each quantified ingredient. If your product contains green tea extract, chamomile, ginseng, or any ingredient with a documented pesticide issue, add a pesticide residue screen.

Step 4: Check your COA formatting. Amazon’s verification team flags COAs missing specific fields even when the analytical results are clean. Every acceptable COA must include: the lab’s full name and ISO 17025 accreditation number, the test methods referenced (USP, AOAC, or equivalent), a unique sample ID, the testing date, declared specifications, and a clear pass/fail determination. Ambiguous formatting causes unnecessary delays.

Step 5: Build ongoing lot-to-lot testing into your SOP. A 2024 COA doesn’t cover 2026 raw material. Botanical raw materials vary between crop seasons, supplier batches, and sourcing regions. Build a standing purchase order with your analytical testing laboratory so that every new lot of incoming raw material receives at least identity and heavy metals screening before it enters your manufacturing process.

DSHEA Compliance vs. Amazon Compliance — They’re Not the Same Thing

This distinction trips up a lot of brands. DSHEA — the Dietary Supplement Health and Education Act of 1994, implemented as cGMP regulations under 21 CFR Part 111 — sets the federal legal minimum for dietary supplement safety and documentation. It requires identity testing on each incoming lot, controls around manufacturing, and proper labeling. It doesn’t specify that testing must be done by an ISO 17025-accredited facility.

Amazon’s program sits on top of DSHEA, not inside it. You can be fully 21 CFR 111-compliant and still fail Amazon’s verification process if your COA methodology or accreditation status doesn’t meet their additional criteria. The cleanest approach is to meet both simultaneously: ISO 17025-accredited analytical testing laboratory results, methods referenced against USP or AOAC standards, COAs formatted with every field their verification team looks for. Getting there isn’t expensive. For a typical botanical supplement with three to five ingredients, a complete dossier covering identity, heavy metals, microbiology, and potency generally runs $600–$1,200 depending on ingredient complexity and method selection — a fraction of the revenue at risk during a listing suspension.

The audit you do now is cheaper than the one Amazon forces on you later.
